چکیده

This study investigated the toxic effects of 2,4-dichlorophenoxyacetic acid on male Sprague-Dawley rats. In which, 30 rats were divided into three even groups. First group received orally normal saline (3 ml/kg b.w.) and kept as normal control in parallel to treated groups (5 rats each), second group received orally 2,4-dichlorophenoxyacetic acid (30 mg/kg b.w.) for 2 months and third group received orally 2,4-dichlorophenoxyacetic acid (30 mg/kg b.w.) for 6 months. At the end of each treatment, specimens from the liver, kidneys, lungs, testicles, epididymis and brain were obtained and subjected for histopathological examination. After 2 months of treatment with 2,4-dichlorophenoxyacetic acid, the overall changes were milder and ranged from vascular to necrobiotic findings. On the other hand, changes seen after 6 months were severe particularly in the liver and kidney. In the liver, there were focal areas of necrosis and preneoplastic changes in the form eosinophilic preneoplastic foci, oval cell proliferation and spongiotic pericytoma. Kidneys showed glomerular swelling, thickening of the glomerular basement membrane and proliferation of the renal tubular epithelium. Lesions in the other organs included necrosis and sloughing of bronchiolar epithelial lining and mononuclear cellular infiltration in the lungs, necrosis of seminiferous tubular epithelium in the testicles and neuronal degeneration in the brain. In conclusion, 2,4-dichlorophenoxyacetic acid induced deleterious pathological effects on the vital organs including preneoplastic changes in the liver in Sprague-Dawley rats.

Sex-dependent differences in the disposition of 2,4-dichlorophenoxyacetic acid in Sprague-Dawley rats, B6C3F1 mice, and Syrian hamsters.

2,4-Dichlorophenoxyacetic acid (2,4-D), a widely used broadleaf herbicide, is under investigation in a study of peroxisome proliferators. To supplement that study, male and female rats, mice, and hamsters were dosed with 14C-2,4-D orally at 5 and 200 mg/kg and tissue distributions were determined. Blood, liver, kidney, muscle, skin, fat, brain, testes, and ovaries were examined. At early time p...

Evaluation of Lead Acetate Side Effects on Rat Hippocampus and the Effects of Vitamin C on These

Purpose: To investigate the effect of ascorbic acid against lead-induced neurotoxicity in the rat hippocampus. Materials and Methods: The heads of 40 male Sprague–Dawley rats were divided into 4 groups: normal, control, lead-treated and lead plus ascorbic acid-treated. Lead acetate (20mg/kg) was administered intraperitonealy to rats for 7days in third and fourth groups. During this period, rats...

Life-Stage-, Sex-, and Dose-Dependent Dietary Toxicokinetics and Relationship to Toxicity of 2,4-Dichlorophenoxyacetic Acid (2,4-D) in Rats: Implications for Toxicity Test Dose Selection, Design, and Interpretation

Life-stage-dependent toxicity and dose-dependent toxicokinetics (TK) were evaluated in Sprague Dawley rats following dietary exposure to 2,4-dichlorophenoxyacetic acid (2,4-D). 2,4-D renal clearance is impacted by dose-dependent saturation of the renal organic anion transporter; thus, this study focused on identifying inflection points of onset of dietary nonlinear TK to inform dose selection d...

Acute and sub-chronic toxicological evaluation of ethanol extract of Solanum trilobatum Linn.

Background and objectives: Solanum trilobatum plant parts such as berries and flowers are normally used for the treatment of respiratory illnesses. The toxicity profile of the plant and its parts are not clear. Hence, the present study was planned to investigate the toxicological effects of ethanol extract of leaves of S. trilobatum (EEST) using acute and sub-...

Effects of pure 2,4-dichlorophenoxyacetic acid on cultured rat embryos.

2,4-dichlorophenoxyacetic acid (2,4-D), a plant growth regulator, has been used worldwide as a herbicide. Previously we evaluated the prenatal developmental effects of 2,4-D by feeding it to pregnant rats and found that it is maternally toxic and embryolethal, and it induces urogenital malformations in rat fetuses. In the study presented here, we investigated the effects of pure 2,4-D on rat em...
